Production of Invertase from Aspergillus Niger Using Fruit Peel Waste as a Substrate
نویسنده
چکیده
Aim of present study was production, purification and characterization of invertase by Aspergillus niger (isolated from soil) using fruit peel waste as a substrate. Screening of various fungi grown on potato dextrose-agar (PDA) was done using liquid medium of 50% sucrose-Cazpek. After incubation at 28 ̊C for 10 days, the invertase activity was determined in the culture filtrate using Fehling’s solution. The positive result was indicated by brown and green precipitate. Sugar generated by the hydrolysis of sucrose was measured by DNSA method. A. niger produced maximum invertase on 4 day of incubation under optimized culture conditions i.e. pH 5.0, temperature 30°C, inoculum size 10 -10 spores/ml in Czapek Dox using fruit peel waste as a substrate by submerged fermentation (SmF). Invertase enzyme was also produced by A. niger from other carbon source (lactose, fructose, sucrose etc.) as substrate. Out of these fructose showed maximum production of invertase. Fruit peel may be proved a good economic source for invertase production on an industrial scale.

Optimization and characterization of invertase by Aspergillus fumigatus using fruit peel waste as substrate
The filamentous fungi A. fumigatus produced high levels of invertase when cultured for 96 hr at 30C and pH 5 in Czapek Dox supplemented with fruit peel waste as substrate. Enhanced production occurred on addition of sucrose and yeast extract as nutritional factors. The crude extract was purified to 4.73 fold with recovery of 4.91% by DEAE-column chromatography and the molecular weight was estim...
متن کاملDate Wastes as Substrate for the Production of a-Amylase and Invertase
Background: In Algeria the date wastes production is estimated at least 85.000 tones. Date wastes is an economic source of carbohydrates for conversion to industrial enzymes because it is readily available and relatively low priced. Objective: The aim of the present study was to investigate the potential of using date wastes as a substrate for the production of a-amylase and invertase. Materia...

متن کاملUtilization of molasses and sugar cane bagasse for production of fungal invertase in solid state fermentation using Aspergillus niger GH1
Agro-industrial wastes have been used as substrate-support in solid state fermentation for enzyme production. Molasses and sugarcane bagasse are by-products of sugar industry and can be employed as substrates for invertase production. Invertase is an important enzyme for sweeteners development. In this study, a xerophilic fungus Aspergillus niger GH1 isolated of the Mexican semi-desert, previou...
